What gutter margin does my KDP book need?
It depends on the final page count of the formatted book, not the manuscript: at least 0.375" up to 150 pages, 0.5" to 300, 0.625" to 500, 0.75" to 700, and 0.875" up to 828 pages, with outside, top and bottom margins of at least 0.25". Because the count that matters is the typeset PDF's, check the exported file — Ceeryo's preflight measures it and applies the right bracket automatically.

Which page count decides the gutter — manuscript or book?+
The typeset book's. A 90,000-word manuscript might be 320 pages at 6×9 in one theme and 350 in another, and the bracket follows the final count — which is why the check runs after formatting.
Do these rules change with trim size?+
The gutter brackets are by page count, not trim. The outside/top/bottom minimum of 0.25" applies across trims; books with bleed add 0.125" to the outside edges.
